Título: Investigating the impact of CX3CR1+ macrophage depletion on gut barrier integrity.

Resumo: The intestinal barrier regulates nutrient absorption and prevents microbial translocation. CX3CR1⁺ macrophages play a critical role in maintaining epithelial integrity and immune homeostasis. This study examines the impact of CX3CR1⁺ macrophage depletion on intestinal epithelial structure, immune composition, and permeability using the CX3CR1-Cre;iDTR model. Flow cytometry revealed Ly6C⁺ monocyte expansion in the small intestine, reduced T cells, increased B220⁺ B cells, and fewer antigen-presenting cells in the colon. ELISA showed elevated serum IgG, suggesting systemic immune activation upon CX3CR1⁺ cell depletion. Histological analysis demonstrated epithelial disruption, goblet cell loss, and diffuse architectural alterations. Tight junction gene expression analysis by RT-qPCR indicated barrier impairment, with occludin, claudin-3, and claudin-4 downregulated in the small intestine, while claudin-2 was upregulated in the colon. These findings underscore CX3CR1⁺ macrophages as essential regulators of mucosal immunity and barrier function. Targeting the CX3CL1/CX3CR1 axis may offer a therapeutic strategy for restoring gut integrity in inflammatory disorders.
Descrição: A barreira intestinal regula a absorção de nutrientes e impede a translocação microbiana. Os macrófagos CX3CR1⁺ desempenham um papel crítico na manutenção da integridade epitelial e da homeostasia imune. Este estudo investiga o impacto da depleção de macrófagos CX3CR1⁺ na estrutura epitelial intestinal, composição imune e permeabilidade, utilizando o modelo transgénico CX3CR1-Cre;iDTR. A citometria de fluxo revelou uma expansão de monócitos Ly6C⁺ no intestino delgado, redução de células T, aumento de células B B220⁺ e menor número de células apresentadoras de antigénio no cólon. A análise por ELISA demonstrou um aumento nos níveis séricos de IgG, indicando ativação imune sistémica, após a depleção de células CX3CR1⁺. A análise histológica evidenciou disrupção da estrutura epitelial, perda de células caliciformes e alterações difusas na arquitetura tecidular. A análise da expressão de genes das tight junctions por RT-qPCR revelou comprometimento da barreira, com redução da ocludina, claudina-3 e claudina-4 no intestino delgado e um aumento da claudina-2 no cólon. Estes achados destacam os macrófagos CX3CR1⁺ como reguladores essenciais da imunidade da mucosa e da função de barreira. O eixo CX3CL1/CX3CR1 pode representar uma estratégia terapêutica promissora para restaurar a integridade intestinal em condições inflamatórias.
