Title: Epigenetic alterations in sperm cells from infertile patients

Abstract: Epigenetic modifications regulate chromatin conformation and transcription factor accessibility to regulatory regions of the genome, controlling gene expression without altering the DNA sequence itself and being stably transmitted throughout cell divisions. One of the most well studied epigenetic marks is DNA methylation, which controls the monoallelic, parental-dependent expression of imprinted genes. Paternal imprinting marks are established in the male germ line, so that the mature gametes - the spermatozoa - transmit correct imprints to the future embryo. Anomalies in the establishment of imprinting marks can affect embryonic and placental development and result in the birth of children carrying imprinting syndromes, such as Silver-Russell and Beckwith-Wiedemann syndromes. Here we review the literature on the observations of imprinting errors in the male gamete, in the context of disturbances in spermatogenesis resulting in male infertility, focusing on the evidence described by our group and others. We provide a clinical perspective on the implementation of sperm methylation analysis as a tool to improve diagnostic and therapeutic strategies in Assisted Reproduction Treatments (ART) and highlight the importance of understanding the molecular mechanisms underlying spermatogenic defects and male infertility.
Description: As modificações epigenéticas regulam a conformação da cromatina e a acessibilidade dos fatores de transcrição a regiões reguladoras do genoma, controlando a expressão génica sem alterar a sequência de DNA em si, sendo transmitidas de forma estável ao longo das divisões celulares. Uma das marcas epigenéticas mais bem estudadas é a metilação do DNA, que controla a expressão monoalélica e dependente da origem parental dos genes sujeitos ao mecanismo de imprinting genómico. As marcas de imprinting paternas são estabelecidas na linha germinativa masculina, de forma que os gametas maduros - os espermatozoides - transmitam as marcas de imprinting corretas para o futuro embrião. Alterações no estabelecimento das marcas de imprinting podem afetar o desenvolvimento embrionário e placentário, podendo resultar no nascimento de crianças com síndromes de imprinting, como as síndromes de Silver-Russell e Beckwith-Wiedemann. Neste artigo, fazemos uma revisão da literatura em relação às descrições de erros de imprinting nos gametas masculinos, no contexto de perturbações da espermatogénese que têm como consequência infertilidade masculina, com foco nas evidências descritas pelo nosso grupo e por outros investigadores. Apresentamos uma perspetiva clínica sobre a implementação da análise de metilação dos espermatozóides como uma ferramenta para melhorar as estratégias diagnósticas e terapêuticas nos tratamentos de Procriação Medicamente Assistida (PMA) e destacamos a importância de compreender os mecanismos moleculares subjacentes a defeitos na espermatogénese e na infertilidade masculina.
