Title: Old-onset chronic alcohol consumption effects on the cholinergic neurons of the pedunculopontine and laterodorsal tegmental nuclei of the rat: an unbiased stereological study.

Abstract: The brain cholinergic system comprises two main recognized subdivisions, the basal forebrain cholinergic nuclei and the brainstem cholinergic nuclei. The effects of aging and chronic alcohol consumption on the basal forebrain cholinergic nuclei have been extensively investigated, but there are few studies that have examined those effects on the brainstem cholinergic nuclei. Furthermore, to the best of our knowledge, no studies have addressed the cumulative effects of aging and late-onset chronic alcohol consumption on the pedunculopontine tegmental (PPT) and laterodorsal tegmental (LDT) nuclei. Thus, in the present study, we have examined, using stereological methods, the effects of long-term prolonged alcohol consumption (6 months) starting at 18 months of age on the neuronal cholinergic population of the PPT and LDT of male Wistar rats. For that purpose, we estimated the total number of PPT and LDT choline acetyltransferase (ChAT)-immunoreactive neurons in all experimental groups, namely, in adult, adult-ethanol treated, old and old-ethanol treated rats. We found that prolonged old-onset chronic alcohol intake did not induce changes in the total number of the LDT cholinergic neurons. Additionally, preliminary data seems to reveal in the PPT cholinergic neurons the same tendency observed in the LDT. These results suggest a remarkable resistance of the brainstem cholinergic neurons compared to other cholinergic cell populations in the brain and highlight the relevance of find all the factors that are putatively involved in the greater resistance of the brainstem cholinergic neurons.
Description: O sistema colinérgico do cérebro compreende duas subdivisões principais, os núcleos colinérgicos do prosencéfalo basal e os núcleos colinérgicos do tronco cerebral. Os efeitos do envelhecimento e do consumo crónico de álcool nos núcleos colinérgicos do prosencéfalo basal têm sido amplamente investigados, mas existem poucos estudos que tenham examinado estes efeitos nos núcleos colinérgicos do tronco cerebral. Além disso, tanto quanto sabemos, nenhum estudo abordou os efeitos cumulativos do envelhecimento e do consumo crónico álcool iniciado numa idade mais avançada nos núcleos pedunculopontino (PPT) e tegmentar laterodorsal (LDT). Assim, no presente estudo, examinámos, através de métodos estereológicos, os efeitos do consumo prolongado de álcool a longo prazo (6 meses), iniciado aos 18 meses de idade, na população neuronal colinérgica do PPT e do LDT em ratos machos Wistar. Para este efeito, estimámos o número total de neurónios do PPT e do LDT imunorreativos para a colina acetiltransferase (ChAT) em todos os grupos experimentais, ou seja, em ratos adultos, adultos tratados com etanol, velhos e velhos tratados com etanol. Verificámos que o consumo crónico prolongado de álcool iniciado numa idade mais avançada não induziu alterações no número total de neurónios colinérgicos do LDT. Além disso, os dados preliminares obtidos nos neurónios colinérgicos do PPT parecem revelar a mesma tendência observada no LDT. Estes resultados sugerem uma resistência notável dos neurónios colinérgicos do tronco cerebral em comparação com outras populações de células colinérgicas do cérebro e realçam a relevância de encontrar todos os fatores que estão potencialmente envolvidos na maior resistência dos neurónios colinérgicos do tronco cerebral.
