| Title: | Effects of chronic alcohol consumption and withdrawal on neuropeptide expression and cholinergic innervation of the rat basolateral amygdala. |

| Abstract: | The basolateral amygdala (BLA), a critical brain structure, receives dense cholinergic innervation from the basal forebrain cholinergic system, and contains neuropeptide Y (NPY) and vasoactive intestinal peptide (VIP) expressing interneurons, both implicated in modulation of various responses and behaviours within the central nervous system. Despite extensive research on the effects of chronic alcohol consumption and withdrawal on the brain, little is known about its impact on the BLA. Thus, in this study we investigated the effects of long-term alcohol ingestion (6 months) and subsequent withdrawal (2 months) on the expression of NPY and VIP and on the cholinergic innervation in the BLA of Wistar male rats. For this purpose, we quantified the BLA density of NPY- and VIP-immunoreactive neurons and vesicular acetylcholine transporter (VAChT)-immunoreactive varicosities. Our results revealed no significant changes in the density of these interneuronal populations or in the cholinergic innervation following chronic ethanol exposure and protracted withdrawal. The present results show that the effects of prolonged alcohol ingestion and protracted withdrawal on the BLA expression of NPY and VIP, as well as on the BLA and cholinergic innervation, differ from those previously observed in other brain regions, revealing a remarkable resistance of the BLA to the deleterious effects of these noxae, regarding the analyzed parameters. |
| Description: | A amígdala basolateral (BLA), uma estrutura cerebral basilar, recebe uma inervação colinérgica densa do sistema colinérgico do prosencéfalo basal, e contém interneurónios que expressam o neuropeptídeo Y (NPY) e o polipeptídeo vasoactivo intestinal (VIP), ambos implicados na modulação de várias respostas e comportamentos no sistema nervoso central. Apesar da extensa investigação sobre os efeitos do consumo crónico de álcool e da abstinência no encéfalo, pouco se sabe sobre o seu impacto na BLA. Assim, neste estudo investigámos os efeitos da ingestão crónica de álcool (6 meses) e da subsequente abstinência (2 meses) na expressão de NPY e VIP e na inervação colinérgica na BLA de ratos machos Wistar. Para o efeito, quantificámos a densidade de neurónios imunorreativos para o NPY e o VIP na BLA e as varicosidades imunorreativas para o transportador vesicular de acetilcolina (VAChT). Os nossos resultados revelaram não haver alterações significativas na densidade destas populações interneuronais ou na inervação colinérgica após a exposição crónica ao etanol e a abstinência alcoólica prolongada. Estes resultados mostram que os efeitos da ingestão prolongada de álcool e da abstinência prolongada sobre a expressão de NPY e VIP, bem como na inervação colinérgica da BLA, diferem de resultados anteriormente observados noutras regiões cerebrais, revelando uma resistência notável da BLA aos efeitos nocivos destas noxas relativamente aos parâmetros analisados. |
