| dc.description | Existe atualmente um aumento significativo de doenças no mundo e como resposta, uma maior quantidade disponível de medicamentos para providenciar uma maior eficácia nos sistemas de saúde. A atual disponibilidade de medicamentos a nível comercial é bastante relevante sendo os seus benefícios bem como a sua importância terapêutica irrefutáveis. Torna-se assim incontestável para a vida humana a importância de novos medicamentos. Existem diversas etapas durante processo de conceção de medicamentos até que este atinja a fase final que corresponde aos testes clínicos. Uma etapa do processo de conceção de medicamentos é testar in vivo o novo medicamento sintetizado. Esta fase caracteriza-se por ser uma fase dispendiosa em termos de custos como também dolorosa para os animais que são submetidos aos testes. Para evitar ou pelo menos reduzir, os testes em animais, as empresas farmacêuticas e os laboratórios de investigação optam por um procedimento designado biopsia onde obtêm uma pequena amostra de tecido maligno(humano ou animal), que é bastante inofensiva para o fornecedor biológico.Com essas amostras, eles podem replicar "ad infinitum" as células em laboratório, onde cada célula replicada dessa maneira é designada de linha celular/ células cancerígenas (cell lines). Dessa forma, podem ser testados novos medicamentos em milhares de doses diferentes em amostras desses tecidos replicados de uma maneira menos dispendiosa e monitorizar seus efeitos bem como a sua eficácia. Estes tecidos replicados também permitem aos investigadores testar novas abordagens e terem um processo adequado para comparação de resultados. O objectivo desta dissertação é a aplicação de Data Mining a dados públicos, já utilizados em um estudo original para o problema de regressão, para conseguir prever a eficácia de medicamentos em linhas celulares. Para além dos algoritmos utilizados no estudo original, diferentes algoritmos foram introduzidos, foi feito uma análise de selecção de características bem como a transformação do problema de regressão em classificação e respectiva análise. | |
| dc.description.abstract | There is currently a significant increase of diseases in the world and, therefore, there is a greater amount of medication available to provide greater effectiveness in health systems. The current availability of medicines at the commercial level is quite relevant, and their benefits and therapeutic importance are irrefutable. Thus, the importance of new medicines is undeniable for human life. There are several stages during the drug design process until it reaches the final phase that corresponds to clinical tests. One step in the drug design process is to test the newly synthesized drug in vivo. This phase is characterized by being an expensive phase in terms of costs as well as painful for the animals that are submitted to the tests. To avoid or at least reduce, animal testing, pharmaceutical companies, and research laboratories opt for a procedure called biopsy where they obtain a small sample of malignant tissue (human or animal), which is quite harmless to the biological supplier. These samples can replicate "ad infinitum" cells in the laboratory where each cell replicated in this way is called a cell line. That way, they can test new drugs in thousands of different doses on samples of these replicated tissues in a less expensive way and monitor their effects as well as their effectiveness. These replicated tissues also allow researchers to test new approaches and have an appropriate process for comparing results. The objective of this dissertation is the application of Data Mining to public data, already used in an original study for the regression problem, that reports the results of the effectiveness of drugs using cell lines. In addition to the algorithms used in the original study, different algorithms were introduced, a feature selection was carried out, as well as the transformation of the regression problem into classification one and respective analysis. | |
