Title: The NFkB signaling pathway in papillomavirus-induced lesions: Friend or foe?

Abstract: Papillomaviruses induce a range of benign andmalignant lesions in their hosts, including cervical cancer, that isassociated with high-risk human papillomavirus (HPV) types. Thenuclear factor kappa-light-chain-enhancer of activated B-cells(NFĸB) plays a pivotal role in HPV-infected cells, and itsexpression and activity are modulated by several viraloncoproteins. NFĸB modulation seems to first facilitate viralpersistence and immune evasion, and later to drive tumourprogression, but the many conflicting results and the complexityof its signaling networks require great prudence while interpretingthe role of NFĸB in papillomaviral lesions. Accordingly, thepharmacological targeting of the NFĸB pathway in HPV-inducedlesions is a complex and currently unmet challenge. This reviewdeals with recent findings concerning NFĸB activation in HPVinfected cells, its role in viral persistence, cell transformation andtumour progression, and with current efforts to target this pathwayfor cancer prevention and therapy.
